Let's start by tuning down to drop D. You simply take the open E string and tune it down one whole step to D. Now the the three lowest strings outline a nice and low open D power chord.

Let's talk a bit about the time signature here; you may have noticed, or felt rather, that the riff is not just a straight 4/4. What we're dealing with here is the time signature 7/4.

But you really don't want to have sit and count all this to try to figure out the rhythm of the riff. What you should do is listen to the riff, over and over again, until the sound and feel of it is locked in your mind. It should be completely second nature to play this as easily as something in 4/4 - you shouldn't sit and count it, you should just be able to play it by heart because you know it that well!
